Bovine × Murine T-Cell Hybridomas Specific for Bovine Herpesvirus 1 (BHV-1) Glycoproteins

摘要: Difficulties in the isolation and long-term maintenance of bovine herpesvirus-1 (BHV-1) specific T-cell clones have hindered analysis cell-mediated immune response to this virus. In an effort identify epitopes virus, murine hybridomas for BHV-1 were generated as alternative clones. Peripheral blood lymphocytes from a calf immunized with restimulated vitro virus generate bulk cultures. The antigen-specific T-cell-enriched culture fused receptor-deficient mutant thymoma cell line BW 5147. screened their ability produce interferon-gamma stimulation. Hybridomas various specificities obtained. One them was glycoprotein gI, two gIV, while three other gIII. hybridoma responded stimulation BHV-1, but not any glycoproteins gIII, or suggesting that proteins than these major may be involved BHV-1. Of hybridomas, one MHC Class I restricted, all others II restricted.
