Control of rRNA synthesis in Escherichia coli at increased rrn gene dosage. Role of guanosine tetraphosphate and ribosome feedback.

摘要: Abstract The effects of extra, plasmid-borne rRNA genes on the synthesis rate in Escherichia coli were examined by measuring fraction total RNA that is and tRNA (rs/rt), cytoplasmic concentration guanosine tetraphosphate (ppGpp), absolute rates protein synthesis. Experiments carried out different growth media with two strains E. coli, B/r K-12. results indicated: 1) increased rrn gene dosage from either intact or defective reduced bacterial ribosome activity (protein rate/average ribosome), rs/rt. 2) Extra intact, but not extra defective, caused level ppGpp to be comparison pBR322-carrying control strain. 3) As a function ppGpp, rs/rt was genes. 4) rate/rrn presence genes; this reduction greater than An analysis these showed they are consistent hypothesis feedback effector role translating ribosomes.
