摘要: Human CD48, a membrane-bound, glycosylphosphatidylinositol (GPI)-linked glycoprotein, is potential tumour target for the treatment of leukaemias and lymphomas. CD48 expressed on T-and B-cells, however < 5% CD34(+) progenitor cells express CD48. A truncated, 45kDa soluble form full length was in Chinese hamster ovary (CHO) cells, shown to consist broad range charge isoforms, with most abundant isoforms between pI 4.5 5.0. The truncated bind antibodies raised against native, GPI-linked by surface plasmon resonance analysis. synthetic, human, scFv immunoglobulin gene library screened recombinant phage display, an antibody fragment, (designated N2A) isolated after four rounds biopanning. N2A reassembled as human IgG1 monoclonal antibody, CHO binding IgG1-N2A confirmed resonance.
