摘要: 5-Lipoxygenase (5-LOX) catalyses two steps in the biosynthesis of leukotrienes (LTs), lipid mediators inflammation derived from arachidonic acid. LTs function normal host defence, and have pathophysiological roles chronic inflammatory diseases, as asthma atherosclerosis. Also, possible effects 5-LOX products relation to tumorigenesis been described. is also involved anti-inflammatory/pro-resolving lipoxins. mainly expressed myeloid lymphoid cells its expression upregulated during cell differentiation. Cellular activity regulated a complex manner depends on cellular localisation, phosphorylation, intracellular calcium concentration redox status cell. Thus, insight regarding biochemistry relevant for better understanding physiology, development pharmacotherapy. Key Concepts: Lipoxygenases oxygenate polyunsaturated fatty acids. 5-LOX produces 5-HPETE, together with other dioxygenases lipoxins. The regulatory C2-like domain binds Ca2+, diacylglycerols phosphatidylcholine regulates catalytic domain. 5-LOX interacts various proteins such FLAP, CLP Dicer. The 5-lipoxygenase pathway part innate immune system. 5-LOX are inflammation, allergic reactions, cardiovascular disease well certain types cancer.
