摘要: The tumour suppressor gene p53 has been found to be mutated or inactivated at high frequency in several common human tumours. We have examined a series of exocrine pancreatic carcinomas for over-expression mutant forms by immunohistochemistry with panel specific antibodies. immunodetectable 13 22 (60%) frozen cancers and seven cell lines. One the antibodies, CM1, recognises formalin-fixed, paraffin-embedded archival material using this reagent we 28 124 (23%) cancers. successfully demonstrated presence point mutations direct sequencing genomic DNA extracted from tissue showing CM1 immunoreactivity. conclude that activation is an important event tumorigenesis antibody can detect proportion cases overexpression pathological material.
