摘要: Responsiveness of drug delivery systems (DDS) against internal and external stimuli attracts wide interest as a mechanism that can provide both site-specific release at the target place feedback regulated rate. Biological environment is quite complex effects intricate medium may have on effectiveness stimulus received certain attention. Differently, impact loaded itself responsiveness DDS has been underestimated. Most drugs are not merely trapped in polymer network, but they effectively interact with some moieties. Nearly all drugs, including therapeutic proteins, ionizable amphiphilic molecules, thus ionic, hydrogen bonding hydrophobic interactions commonly exploited to increase loading yield. If moiety involved binding also responsible for (or least partially in) responsiveness, strong behavior be expected. This review gathers relevant examples how modify sensitiveness (stimulus threshold) (actuation) therapeutically stimulus, aims shed light different modes swollen collapsed states, which turn patterns. The information evidences trigger phase transitions hydrogels non-intended drug-responsive (i.e., priori analyte-responsive networks). A better knowledge about effect required step forward clinical application smart unveil novel uses stimuli-responsive DDS.
