A novel core–shell chronotherapeutic system for the oral administration of ketoprofen

摘要: Abstract The aim of a chronotherapeutic medicine is to tailor drug in vivo availability circadian rhythms diseases in order reduce side effects and improve patient compliance. To counteract morning symptoms early pathologies, delivery system able delay ketoprofen release after oral administration has been proposed. Particles made pectin matrix were produced by means prilling; core–shell systems covering the particles with Eudragit S100 ® . developed, containing different drug/polymer ratios, fully characterized terms content, efficiency encapsulation, morphology kinetics. results indicated that loading larger amounts feed solution led spherical mono-disperse beads, high encapsulation reduced gastric simulated fluids (22%). Finally, further (7.3% released fluid) was achieved applying drug/pectin core shell (40% w/w) gastro-resistant polymer. This platform exploits characteristics two selected polymers, i.e. pH dependent solubility peculiarities pectin, degraded slowly alkaline environment selectively intestinal microflora in vivo.
