摘要: Lumican (LUM) is a candidate gene for myopia in the MYP3 locus. In this study, mutant lumican (L199P) transgenic mouse model was established to investigate axial length changes and ultrastructural features of sclera. The by pronuclear microinjection. Transgenic mice wild-type B6 were killed at eight weeks age. Gene expression levels LUM collagen type I (COL1) sclera analyzed quantitative real-time polymerase chain reaction (qPCR), protein assessed Western blot analysis. Ocular lengths measured on enucleated whole eye under dissecting microscope. Ultrastructural fibrils examined with transmission electron microscopy (TEM). both elevated transcriptional levels. mean eyes significantly longer than that (3,231.0 ± 11.2 μm (transgenic group) vs 3,199.7 11.1 (controls), p<0.05 =). Some observed TEM, such as evident lamellar disorganizations abnormal inter-fibril spacing. average fibril diameter smaller their counterparts. These results indicate ectopic may induce enlargement structures distributions This can be used mechanistic study myopia.
