A poliomyelitis model through mucosal infection in transgenic mice bearing human poliovirus receptor, TgPVR21

摘要: Transgenic mice bearing the human poliovirus receptor (TgPVR) are less susceptible to oral inoculation, although they parenteral inoculation. We investigated susceptibility of TgPVR 21 line [Arch. Virol. 130 (1994) 351] through various mucosal routes. Intranasal inoculation a neurovirulent Mahoney strain (OM1) caused flaccid paralysis with viral replication in central nervous system at dose 10(6) cell culture infectious (CCID50), contrast, no following or intragastric same dose. vaccine strain, Sabin 1, CCID50, resulted paralysis. Initial nasal cavity was confirmed by virus isolation and detection negative-stranded replicative intermediates RT-PCR antigens using high-sensitive immunohistochemistry genome/transcripts situ hybridization. Poliovirus-specific IgG antibodies were elevated sera surviving TgPVR21. This model can be used as infection for differentiation attenuated strains.
