The role of intracellular zinc in modulation of life and death of Hep-2 cells.

摘要: Varying intracellular concentrations of zinc in laryngeal Hep-2 cells relation to changing cultivation conditions in vitro were determined by atomic absorption spectrophotometry. Upon standard DMEM with 10% serum, the mean concentration was at 0.88 ± 0.09 μg/mg protein, substantially decreased values exposed a low-serum medium. Next, study effects series physiological and supraphysiological ZnSO4 on their correlation performed. It found that above 100 μM toxic cells, inducing cell death interval 96 h as videomicroscopy, selective nuclear staining, immunofluorescence detection caspase-3 specific cytokeratin 18 fragment. Both types observed, apoptosis being induced moderately 150 μM necrosis higher 300 μM 750 μM, respectively. Lower (1.5–100 μM), other hand, did not produce any measurable changes morphology function same time interval. Zinc 1.5 μM slightly enhance proliferation up certain point, which seemed correlate maximal tolerable momentary level zinc. These results illustrate importance determining levels when trying characterize effect exogenous life cells.
