摘要: Rolling circle replication of a padlock primer is inhibited when it hybridised to target nucleic acid that long or circular. The invention provides methods addressing this problem including cutting the near preferably at site which hybridises with probe, whereby 3′-end cut acts as for rolling probe. Also included method assaying polyepitopic by use two affinity probes each carrying an oligonucleotide tag and probe in association probes.
