摘要: In vitro and animal studies suggest that high concentrations of recombinant human erythropoietin (rHuEPO) might divert multipotent progenitors into erythroid maturation at the expense granulocyte production. We determined whether changes number lineage commitment peripheral blood progenitor cells occur in premature infants during therapy with rHuEPO. Thirty preterm were randomly assigned either to receive 300 IU eopoetin alpha s.c. per kilogram body weight three times a week for four weeks or control group. At study entry after two treatment numbers circulating BFU-E, granulocyte-macrophage colony-forming units (CFU-GM) granulocyte-erythrocyte-macrophage-megakaryocyte CFU (CFU-GEMM) analyzed by semisolid culture technique, CD34+ early myeloid CD34+CD45RA- flow cytometry. As compared group, rHuEPO did not exert any significant modulatory effect on CFU-GM, nor was there change CFU-GEMM, total-CFU, percentage cells. Mean neutrophil count significantly reduced period study. Compared receiving had higher hematocrit values (p = 0.003) absolute reticulocyte counts < 0.001). The median cumulative volume transfused day 0.86 ml (first quartile 0.5 ml; third 1.1 ml) group 0 0.47 0.038). conclude using relatively dose infants, no vivo could be detected.
