摘要: Publisher Summary This chapter elaborates the measurement and preparation of glutamate-oxaloacetate transaminase (GOT). GOT has been detected in micro-organisms all human animal tissues. In humans richest source is heart muscle, followed by brain, liver, gastric mucosa, adipose tissue, skeletal kidney, finally serum with substantially smaller amounts. Several possibilities exist for activity which includes paper chromatography substrates or reaction products after incubation spectrophotometric oxaloacetate formation at 280 mμ. The most widely used methods involve formed from aspartate α-oxoglutarate: one them an enzymatic indicator malic dehydrogenase reduced diphosphopyridine nucleotide. pyridoxal phosphate required as a coenzyme present sufficient quantities tissue samples. Animal organs, especially brain D are rich glutamic dehydrogenase. It necessary to determine amount interference due this enzyme, occurs spite use ammonia-free reagents phosphate–aspartate buffer substituted containing α-oxoglutarate started L -aspartate measurements activity.
