Mouse Connexin 45: Genomic Cloning and Exon Usage

摘要: Connexin 45 is a gap junction protein that prominent in early embryos and widely expressed many mature cell types. To elucidate its gene structure, expression, regulation, we isolated mouse Cx45 genomic clones. Alignment of the DNA cDNA sequences revealed presence three exons two introns. The first contained only 5′ untranslated sequences, while exon 3 remaining UTR, entire coding region, 3′ UTR. An RT-PCR with exon-specific primers was utilized to examine usage F9 embryonal carcinoma cells adult tissues. In all samples, PCR products amplified using 2/exon or 3/exon primer pairs were much more abundant than produced 1/exon 2 pairs, suggesting mRNAs containing 1 relatively rare compared other exons.
