摘要: We previously described a system for cell culture under media perfusion, which was developed for longitudinal measurement of cell-autonomous circadian rhythms in the same population of cells, reported bioluminescently, cited by a recent article in this journal (Fustin et al. 2019), and fully described in Crosby et al. 2017. Briefly, the system consists of an enclosed channel slide, into which adherent cells are seeded, situated within a humidified tissue culture incubator (5% CO2, 37 C). A modified Dulbecco’s Modified Eagle Medium, which features a more physiological glucose level (5 mM) and lower serum concentrations (2%) than commonly employed with adherent cell culture, is pumped across the slide at a rate of 50 μl/hr using a syringe pump, with ETFE tubing of 1 mm internal diameter connecting the media-containing syringe to the cell-containing slide.
