摘要: In this research albumin nanocapsules were fabricated as paclitaxel carrier to the tumor cells. Adsorption of albumin onto nanosilica particles carried out in aqueous media. In order to optimize the fabrication of nanoparticles the Taguchi metod was applied and effect of some parameters such as pH, glutaraldehyde concentration as a cross linking agent, agitation speed and BSA concentration onto the thickness of the albumin layer was considered. Surface of silica nanoparticles was modified by Aminopropyl triethoxy silane (APTES) and adsorption of albumin onto the surface was carried out. Physical properties, morphology and the thickness of the nanoparticles was studied using ultraviolet spectroscopy and Thermogravimetry analyses besides scaning and transmission microscopy. The results indicated that albumin formed a coating layer on the silica and nanoparticles possess core/shell and spherical structure with average diameter of 20 nm. Subsequently, different ratio of drug/nanoparticle was applied in order to loading paclitaxel onto nanoparticles and in vitro release of paclitaxel were considered by using dialyse bag and accumulative drug release pattern was determined utilizing high performance liquid chromatography and optimized value of drug/nanoparticle was obtained 10% and results showed that Higuchi release kinetics model is more compatible whit the experimental data. The in vitro cytotoxicity activity of paclitaxel loaded nanoparticles was assessed using lung cancer cell lines.
