摘要: To understand the secretion and processing of interleukin-1 (IL-1), a Chinese hamster fibroblast cell line (R1610) was transfected with human IL-1 beta cDNA under control SV40 early promoter linked to gene for neomycin resistance. After selecting cells resistant G418, two clones were found constitutively express 31-kD precursor which almost exclusively located in cytosol. Pulse-chase experiments failed show any very little activity detectable supernatants. Furthermore, surface membrane could not be detected, although low levels released upon brief trypsin treatment. Therefore, unlike monocytes, these lack mechanism secreting beta.
