DNA hypomethylation of karyoplasts for bovine nuclear transplantation.

摘要: The objective of this research was to evaluate if DNA hypomethylation in cells used as karyoplasts would improve development bovine nuclear transplantation (NT) embryos. from serum-fed (SF), serum-starved (SS), and 1, or 5 microM 5-azacytidine (5-aza-CR) treated digested with a methylation sensitive enzyme, evaluated for methylation. A significant reduction observed cultured 48 72 hr SS medium well the presence 5-aza-CR when compared SF medium. All other comparisons contained no differences controls. When donor were 5-aza-CR, SF, treatment media hr, difference (P = 0.06) blastocyst rates after NT. One embryo produced by established pregnancy. Four pregnancies resulted embryos cell NT 3 Supplementation culture not beneficial increasing rate establishing pregnancy
