摘要: Publisher Summary This chapter discusses the light scattering/size exclusion chromatography method to study stoichiometry of a protein–protein complex. Interactions protein with itself or other macromolecules play important roles in diverse biological processes. These interactions are characterized by their affinity and stoichiometry. The size (SEC) is used estimate molecular weight complex therefore determine During formation complex, binding an additional may not affect overall hydrodynamic size, conversely, it change carbohydrates' conformation that influences elution position. from scattering measurement independent position reflects only polypeptide if extinction coefficient alone analysis. characteristics make combination SEC easy, accurate, reliable technique. focuses on determination complexes containing at least one glycosylated protein.
