Role of Rac1 and Oxygen Radicals in Collagenase-1 Expression Induced by Cell Shape Change

摘要: Integrin-mediated reorganization of cell shape leads to an altered cellular phenotype. Disruption the actin cytoskeleton, initiated by binding soluble antibody alpha5beta1 integrin, led increased expression collagenase-1 gene in rabbit synovial fibroblasts. Activation guanosine triphosphate-binding protein Rac1, which was downstream necessary for this process, and activated Rac1 sufficient increase collagenase-1. activation generated reactive oxygen species that were essential nuclear factor kappa B-dependent transcriptional regulation interleukin-1alpha, which, autocrine manner, induced expression. Remodeling extracellular matrix consequent alterations integrin-mediated adhesion cytoarchitecture are central development, wound healing, inflammation, malignant disease. The resulting may lead morphogenesis, migration, invasion.
