Novel fabrication of PCL porous beads for use as an injectable cell carrier system

摘要: Injectable polycaprolactone (PCL) porous beads were fabricated for use as cell carriers by a novel isolated particle-melting method (for nonporous beads) and the following melt-molding particulate-leaching beads). The prepared showed highly uniform pore structures with almost same surface interior porosities (porosity, over 90%). PCL (bead size, 400-550 microm) different sizes (25-50 50-100 compared their in vitro (human chondrocyte) growth behavior beads. higher seeding density than size effect between was not significant up to 7 days, but after that time of microm significantly those 25-50 microm. To evaluate tissue compatibility beads, dispersed, uniformly, cold Pluronic F127 solution injected into hairless mice, subcutaneously, gel state at room temperature, leading homogeneous bead delivery. histological findings confirmed are biocompatible: surrounding tissues gradually infiltrated 4 weeks little inflammatory response. this study can be widely applicable carriers.
