Prevention of postsurgical tissue adhesion by anti‐inflammatory drug‐loaded pluronic mixtures with sol–gel transition behavior

摘要: Sol-gel transition temperature-controllable Pluronic F127/F68 mixtures including mildly crosslinked alginate and nonsteroidal anti-inflammatory drug (ibuprofen) were prepared to evaluate their potential as tissue adhesion barrier gels. The sol-gel temperatures of the could be controlled by adjusting ratio polymer concentration. with still flow property provided residence stability mixture gels in body. Ibuprofen was loaded reduce inflammatory response body and, thus, prevent adhesion. gelation not affected but lowered addition ibuprofen. vitro release behavior vivo peritoneal just below investigated. from ibuprofen (1 wt%)-loaded at 37 degrees C examined using a membrane-less dissolution model. released continuously up about 45-65% total loading amount during first 7 days. For evaluation anti-adhesion potential, with/without coated on wall defects rats extents reactions (inflammatory response, granulation formation, toxicity organs) compared. It observed that has positive effect for anti-adhesion. F127/F68/alginate/ibuprofen gel (25 wt% [7/3], 1 ibuprofen) highly effective prevention showed relatively low non-toxicity, thus can good candidate material coatable or injectable gel.
