In Vitro Regeneration of Curcuma caesia Plantlets from Basal Part and via Somatic Embryogenesis

摘要: Plantlets of Curcuma caesia were produced in vitro from newly sprouting vegetative buds tubers. Segments the plantlets the junction between root and basal portion stem were subsequently used as explants to investigate factors affecting callus induction plant regeneration via somatic embryogenesis. The placed on Woody Plant Medium (WPM) together with different concentrations 2,4-dichlorophenoxyacetic acid(2,4-D) and benzyl aminopurine (BAP) in presence light. growth medium supplemented with 5 mg/L BAP 2 2,4-D promoted callus induction after 70 days culture. Sub-culturing same medium enhanced production of friable callus. Culture media containing higher agar promoted the development green somatic embryos Respond somatic embryogenesis was most successful MS 6.0 g/L supplemented with 0.2 whereby developed into green somatic an efficiency 53%. This culture also produced the largest number plantlets.
