TIPE2, a Negative Regulator of Innate and Adaptive Immunity that Maintains Immune Homeostasis

摘要: Immune homeostasis is essential for the normal functioning of immune system, and its breakdown leads to fatal inflammatory diseases. We report here identification a member tumor necrosis factor-α-induced protein-8 (TNFAIP8) family, designated TIPE2, that required maintaining homeostasis. TIPE2 preferentially expressed in lymphoid tissues, deletion mice multiorgan inflammation, splenomegaly, premature death. TIPE2-deficient animals are hypersensitive septic shock, cells hyper-responsive Toll-like receptor (TLR) T cell (TCR) activation. Importantly, binds caspase-8 inhibits activating protein-1 nuclear factor-κB activation while promoting Fas-induced apoptosis. Inhibiting significantly blocks hyper-responsiveness cells.
