摘要: According to the French paradox, red wine consumption reduces incidence of vascular diseases even in presence highly saturated fatty acid intake. This phenomenon is widely attributed phytoalexin resveratrol, a ingredient. Experimental studies suggesting that resveratrol has neuroprotective properties mostly used prophylactic delivery strategies associated with short observation periods. These did not allow conclusions be made about resveratrol's therapeutic efficacy post-stroke. Herein, we systematically analyzed effects prophylactic, acute and post-acute (50mg/kg) on neurological recovery, tissue survival, angioneurogenesis after focal cerebral ischemia induced by intraluminal middle artery occlusion mice. Over an period four weeks, only prolonged sustained recovery as assessed rota rod, tight rope corner turn tests. Although reduced infarct volume enhanced blood-brain-barrier integrity at 2 days post-ischemia elevating downstream signal sirtuin-1, increasing cell survival signals (phosphorylated Akt, heme oxygenase-1, Bcl-2) decreasing death (Bax, activated caspase-3), reduction size day 28 was observed any three experimental conditions. Instead, angiogenesis neurogenesis were noted animals receiving delivery, which elevated concentrations GDNF VEGF brain. Thus, depends successful brain remodeling rather than structural neuroprotection. The promoting effect delayed opens promising perspectives for stroke therapy.
