Determination of therapeutics with growth‐hormone secretagogue activity in human urine for doping control purposes

摘要: The administration of growth-promoting agents such as human growth hormone well compounds with respective secretagogue activity is prohibited in sports according to the regulations World Anti-Doping Agency. Acetylcholine esterase inhibitors have been demonstrated stimulate growth-hormone secretion elderly humans, and new orally active drugs developed provide alternatives therapeutic injections preparations. Preventive anti-doping strategies include method development for emerging potentially misused compounds. Hence, mass spectrometric dissociation behavior three acetylcholine (donepezil, galantamine rivastigmine) a structural analogue SM-130686 were studied using high-resolution/high-accuracy orbitrap spectrometry. These data provided substantial information screening procedures, complementing common methods drug testing. Using liquid-liquid extraction subsequent liquid chromatography/tandem spectrometry (LC/MS/MS) analysis, four target analytes determined at urinary concentrations 15-20 ng/mL, recoveries ranged from 55-97%, assay precisions calculated 5.2-15.8% (intraday) 10.2-21.6% (interday) all applicability authentic urine specimens was tested two study samples after application Reminyl (galantamine) Aricept (donepezil). In both cases, administered desmethylated metabolites detected.
