摘要: OBJECTIVE To investigate the inhibitory effect of polysaccharide Atractylodes macrocephala (PAM) on proliferation and invasion hepatocellular carcinoma cells underlying mechanism. METHODS Hepatocellular HepG2 were treated with different concentrations PAM, their invasive ability examined using CCK-8 assay Transwell assay. Immunofluorescence was performed to detect expression level β-catenin, real-time PCR Western blotting used mRNA protein expressions AKT, GSK-3β MMP-2 in cells. The changes proliferation, invasiveness pGSK-3β MMP2 following treatment LiCl/PAM/LiCl plus PAM. RESULTS PAM significantly reduced cell viability, number migration cells, levels β-catenin (P < 0.05), obviously inhibited phosphorylation AKT 0.05) a dose-dependent manner. rescue experiment showed that LiCl reversed inhibition invasiveness, Wnt/β-catenin pathway induced by CONCLUSIONS can inhibit vitro possibly inhibiting signaling pathway.
