摘要: SUMMARY Anti-haemoglobin S antibodies were raised in horses and purified by affinity chromatography. These recognize β6 valine, while they fail to bind haemoglobins with a glutamyl (Hb A) or lysyl C) residue this position. The anti-Hb composed of equine IgG(a, b) IgG(T) subclasses failed cause precipitation Hb S, evidently because the bivalency both antigen. Identification single erythrocytes was achieved reacting fluorescein isothiocyanate-conjugated haemoglobin antigen fixed blood smears. All red cells from persons sickle cell trait anaemia are labelled S-FITC, normal other mutant genotypes not. Erythrocytes new-borns antibody, although intensity labelling varies cell. This method cellular identification preserves morphological characteristics reactive permits observations pattern synthesis among various degrees maturity. Fluorescent smears may be applicable prenatal diagnosis haemoglobinopathies.
