Analysis of the mammalian gadd45 gene and its response to DNA damage

摘要: The gadd45 gene is transcriptionally activated through at least two different mechanisms; one following treatment with base-damaging agents such as methylmethane sulfonate and UV radiation the other ionizing radiation. To investigate sequences involved in induction of by producing high levels base damage, hamster, human, mouse genes were sequenced. Comparison these revealed a level conservation between species 1500 pairs proximal promoter 700 within third intron. However, regions, there was no any transcription factor binding sites known to confer DNA damage responsiveness. hamster inducible both rodent human cell lines line. This indicates that sequence elements factors are conserved mammalian cells. Deletion analysis did not reveal specific which conferred inducibility maximal response required large portion promoter. radiation, suggesting outside region used, p53 site intron, necessary. GADD45 mapped chromosome 1p31.1-31.2.
