摘要: Gene targeting in embryonic stem (ES) cells allows the production of mice with specified genetic mutations. Currently, germline-competent ES cell lines are available from only a limited number mouse strains, and inappropriate cell/host blastocyst combinations often restrict efficient gene-targeted mice. Here, we describe derivation C57BL/6J (B6) compare effectiveness two host donors, FVB/NJ (FVB) coisogenic strain C57BL/6-Tyr(c)-2J (c2J), for germline chimeras. We found that when B6 were injected into c2J blastocysts, high rate coat-color chimerism was detected, transmission could be obtained few injections. In all but one case, highly chimeric transmitted to 100% their offspring.
