摘要: Abstract WH1fungin, a surfactin cyclopeptide from Bacillus amyloliquefaciens WH1, is firstly reported as novel immunoadjuvant, which can markedly enhance the immune response when given in mixture with antigens. After intramuscular or subcutaneous immunization, WH1fungin help to induce both of durable humoral and cellular response, even strong Freund's adjuvant. Both IgG1 IgG2a antigen-specific antibodies were elicited immunizations indicating mixed Th1/Th2 response. Splenocytes mice intramuscularly immunized OVA plus responded CTL peptide stimulation resulting an increase CD8 + TNF-α IFN-γ T cell populations, also CD4 cells populations was found subcutaneously Th stimulation. These results further suggest that helps elicit responses OVA. The potential mechanism immunoadjuvant investigated. In vitro assays showed could enter into RAW 264.7 cells, ROS accumulation, expression surface markers cytokines cells. Further investigation suggested might exert its adjuvant activity by ligating TLR-2 antigen present such 264.7. Taken together, very potent for development vaccines future.
