摘要: A cDNA clone, designated PsDof1, encoding a novel member of the Dof DNA-binding protein family was isolated from pea. Random-binding-site selection and gel retardation experiments showed that GST-PsDof1 recombinant bound to specific DNA sequences with an AAAG core. metal-chelating agent, 1, 10-phenanthroline, abolished binding DNA, but addition Zn2+ restored it, indicating zinc ions are required for its activity. Interestingly, there possible sites PsDof1 in promoter sequence. Indeed, binds AAAG-containing sequence gene higher affinity. These results suggest is transcription factor involved own transcriptional regulation.
