摘要: Cis-regulatory elements involved in the activation of plant defense-related gene encoding chalcone synthase 1 (PsChs1) pea (Pisum sativum L.) were examined by transient transfection, gel mobility shift assay and vitro DNase I-footprinting analysis. Transient transfection revealed that a 61 bp DNA fragment spanning from -242 to -182 PsChs1 was required for maximal promoter activity possibly enhancement elicitor-mediated activation. Nuclear isolate elicitor-treated epicotyl tissues contained some factor(s) specifically bound this form complex with low (LMC, complex) assay. analysis LMC among three protected regions detected fragment, two identical AT-rich sequence, TAAAATACT. Site directed mutation either or both sequences, TAAAATACT TGGAATACT, resulted reduction loss ability LMC. Detailed demonstrated region -226 containing promoter-distal motif imperative PsChs1.
