摘要: BACKGROUND: Quality control of platelet (PLT) concentrates is challenging, due to PLT lesions, which are difficult detect with routine methods. The search for reliable lesion biomarkers focused on the role PLTs in primary hemostasis. transfusions also have a significant impact secondary In this phase, responsiveness small amounts thrombin crucial. PAR1 and PAR4 protease-activated receptors responsible reactivity human PLTs. This study should elucidate if levels those two changing during storage changes an aggregation support generation. STUDY DESIGN AND METHODS: from buffy coat preparations were stored SSP+ solution 9 days at 22 ± 2°C horizontal flatbed agitator, samples taken daily analysis. evaluated using Western blot was measured Born aggregometry specific or agonists. Thrombin generation calibrated automated thrombography. RESULTS: Levels both (PAR1 PAR4) started decrease after 5 storage. PAR1-mediated remained constant, whereas PAR4-mediated decreased time. Rate accelerated storage. CONCLUSION: Decreasing PARs influenced PAR4-mediated, but not PAR1-mediated, aggregation. senescent increased, may be attributed other mechanisms promoting increased phosphatidylserine exposure.
