摘要: Jaagsiekte sheep retrovirus (JSRV) is an exogenous of that induces a contagious lung cancer, ovine pulmonary adenocarcinoma (OPA). JSRV potent carcinogen in the experimental setting, inducing end-stage tumors at around 6 weeks age when newborn lambs are inoculated intratracheally. Despite this rapid oncogenesis, inspection genome sequence does not reveal any obvious viral oncogenes. In review, recent advances studies oncogenic transformation described. Molecular cloning infectious and provirus was instrumental studies. DNA transfection proviral into mouse NIH3T3 cells results morphological transformation, indicating carries oncogene. Further experiments identified envelope protein as transforming gene, PI3 kinase docking site cytoplasmic tail transmembrane (TM) shown to be necessary for transformation. Avian DF-1 infected with avian retroviral vector (RCAS) expressing also undergo tumorigenic Possible mechanisms discussed, cooperating role insertional activation proto-oncogenes tumorigenesis considered. The potential may infection replication vivo.
