The neuroprotective effect of lithium against high dose methylphenidate: Possible role of BDNF.

摘要: Methylphenidate (MPH) is a neural stimulant with unclear neurochemical and behavioral effects. Lithium neuroprotective agent in use clinically for the management of manic-depressive other neurodegenerative disorders. This study investigated protective effect lithium on MPH-induced oxidative stress, anxiety, depression cognition impairment. Forty-eight adult male rats were divided randomly equally into 6 groups. Treatment groups received MPH (10mg/kg) various doses (75, 150 300mg/kg) simultaneously also (150mg/kg) alone 21 days. Elevated Plus Maze Forced Swim Test used to determine level anxiety animals. Morris Water was evaluate spatial learning memory. The hippocampi isolated activity oxidative, anti-oxidant inflammatory factors measured. Also brain derived neurotropic factor expression measured by RT-PCR western blotting. caused behaviors indicative depression-like phenotypes EPM FST impairment MWM. While all mentioned inhibited these significantly increased lipid peroxidation, mitochondrial GSH content IL-1β TNF-α levels hippocampal cells. Moreover superoxide dismutase glutathione peroxidase activities BDNF remarkably decreased. Various attenuated effects mitigated damage, inflammation level. has potential act as against induced toxicity rat this might be mediated hippocampus rats.
