High pressure disruption: a two-step treatment for selective extraction of intracellular components from the microalga Porphyridium cruentum

摘要: The microalga Porphyridium cruentum is known to produce many components of interest. One them B-Phycoerythrin (B-PE), a water-soluble intracellular pigment used as an immunofluorescent probe. Current methods extract this molecule involve total cell disruption and lead mix all the components. Subsequently, purification very complex. An alternative approach B-PE selectively thus simplify procedure has been developed using high-pressure disrupter. Different pressures (from 27 270 MPa), extracting mediums (distilled water original microalgae culture medium), numbers passages (1 3) have tested. Proteins are more extracted than at low pressure in medium. It possible remove part proteins first step then recover enriched fraction higher distilled water.
