Effect of chronic treatment with a purified flavonoid fraction on inflammatory granuloma in the rat. Study of prostaglandin E2 and F2 alpha and thromboxane B2 release and histological changes.

摘要: Abstract S-5682 (Daflon-500 mg), a purified flavonoid fraction, consisting of 90% diosmin (a flavone derivative) and 10% hesperidin flavanone derivative), was administered to rats by intubation in the daily dose 100 mg/d. 15 days after start treatment, polyurethane sponges were implanted subcutaneous connective tissue dorsolumbar region under rapid ether anaesthesia. Similar fragments sponge group control animals who received vehicle (saccharose syrup) only, also oral route. The sacrificed fractions 7 drawn from each two groups (control treated) 4, 8, 16 30 (only 5 on day 30) implantation sponges. granulomas formed removed, weighed their prostaglandin (PG)E2, PGF2 alpha thromboxane (Tx)B2 contents determined. In addition full cell count (polymorphs, lymphocytes, macrophages, plasmocytes giant cells) performed histologically examined. results show that treatment with had following effects: 1. A significant fall mean weight 4 8 observed, reflecting inhibition oedema formation during early phase inflammatory reaction. 2. synthesis PGE2 (78.5% 4) (45.2% 16) inhibited. 3. There very TxB2 (59.5% 4). 4. later reduction migration towards focus occurred which statistically (49.6% total number migrant cells). 5. Multiple histological aspects acute reaction (diapedesis polymorphs, histiocytes macrophages) features chronic (newly microvascularisation granuloma tissue, perivascular oedema, presence collagen fibres) improved.
