Biochemical analysis of the response in rat bone marrow cell cultures to mechanical stimulation

摘要: Bone marrow cells obtained from rat femora were subjected to primary culture with 15% fetal bovine serum in the presence of 10(-8) M dexamethasone, and following trypsin treatment 5 days later seeded on Petriperm dishes which have a flexible bottom. After 2-day subculture, cyclic stress consisting 1 s stretch (0.3% strain. 0.5 Hz) relaxation for 30 min every day was started. Culture tissue removed 2 subculture (immediately prior start stimulation), then 8 (3 6 after stimulation, respectively), at times dry weight, DNA, alkaline phosphatase (ALP) activity, bone Gla protein (BGP, osteocalcin) measured. Both weight DNA showed significant increase stimulated group by 8, while ALP activity 5. The BGP began contrast control it only increased 8. These results support contention that mechanical stimulation promotes differentiation osteogenic enhances formation. Since this experimental model acceleration formation can be reproduced vitro, is extremely useful investigating mechanisms underlying stimulation.
