THE VERTEBRATE GLFG NUCLEOPORIN, NUP98, IS AN ESSENTIAL COMPONENT OF MULTIPLE RNA EXPORT PATHWAYS

摘要: The 97-kD O-linked glycoprotein, Nup98, is a component of the Xenopus laevis nuclear pore complex and only vertebrate GLFG nucleoporin identified (Powers, M.A., C. Macauley, F. Masiarz, D.J. Forbes. 1995. J. Cell Biol. 128:721–736). We have investigated possible roles xNup98 in nucleocytoplasmic transport proteins RNAs by analyzing consequences injecting monospecific polyclonal antibodies to into X. oocytes. show here that injection anti-xNup98 inhibited export multiple classes RNAs, including snRNAs, 5S RNA, large ribosomal mRNA. In contrast, tRNA was unaffected. Injection antixNup98 oocyte cytoplasm had no effect on any RNAs. Significantly, did not inhibit import either karyophilic or snRNPs. This latter result agreement with our previous finding Nup98 an essential element protein pathway. Thus, plays role specifically RNA from nucleus, it appears be pathways.
