Presence of a DNA-4236 bp deletion and 8-hydroxy-deoxyguanosine in mouse cardiac mitochondrial DNA during aging.

摘要: A particular mitochondrial DNA (mtDNA) deletion, the so-called “common deletion”, accumulates progressively with age in human post-mitotic cells. We investigated presence of age-related mtDNA deletions mouse heart and, according to free-radical theory aging, potential involvement reactive oxygen species (ROS). Hearts from young, adult and old male Balb/c mice were homogenized centrifuged order discard nuclear DNA. The supernatant was then utilized prepare by SDS-proteinase K digestion. a mtDNA4236 deletion estimated PCR analysis, separating amplificated segment on agarose gel. incidence 16%, 28% 78% mice, respectively. 8-hydroxy-2′-deoxyguanosine (8-OH-dG), marker oxidation, also determined HPLC-electrochemical analysis. 8-OH-dG not detectable young while its concentrations (moles / 106 moles dG; mean±SD) 59.0±1.41 31.0±4.24 (p<0.02) These data indicate that mtD-NA4236 is associated aging hearts, oxidative damage greater middle-aged than senescent animals.
