A Morphometric Study of Dedifferentiated and Involutional Dark Keratinocytes in 12-O-Tetradecanoylphorbol-13-acetate-treated Mouse Epidermis

摘要: The epidermis of mice treated with 12- O -tetradecanoylphorbol-13-acetate (TPA) was investigated using the method stereological cytology. These procedures were carried out in order to quantify morphological changes taking place basal cells, presumed target cells TPA epidermis. layer composed TPA-induced dark keratinocytes (DC) and clear keratinocytes. DC could be divided into two types. Type 1 characterized by a relatively high volume fraction ribosome nonbundled filaments low bundled filaments. characteristics type I constant over observation period. II density mitochrondria, filaments, cytoplasmic vacuoles. This seldom seen normal but constituted approximately 20% 24 48 hr after treatment TPA. In contrast, exhibited time-dependent such as an increase relative nuclei; mitochondria, ribosomes; decrease Both qualitative quantitative analyses TPA-treated fetal suggest close similarity between epidermis, thus supporting view that are poorly differentiated or dedifferentiated different from involutional DC.
