The non-muscle myosin phosphatase PP1β (flapwing) negatively regulates JNK in wing imaginal discs of Drosophila

摘要: Drosophila flapwing (flw) codes for serine/threonine protein phosphatase type 1β (PP1β). Regulation of non-muscle myosin activity is the single essential flw function that nonredundant with three closely related PP1α genes. Flw thought to dephosphorylate nonmuscle regulatory light chain, Spaghetti Squash (Sqh); this inactivates heavy Zipper (Zip). Thus, strong mutants lead hyperphosphorylation Sqh and hyperactivation activity. Here, we show genetically a Jun Nterminal kinase (JNK) mutant suppresses semi-lethality allele. Alleles JNK puckered (puc) enhance weak allele flw, leading severe wing defects. Introducing binding subunit (Mbs) further enhances genetic interaction lethality. We puc expression upregulated in imaginal discs puc, upregulation modified by Zip. The level phosphorylated (active) elevated enhanced puc. Together, disruption activates
