Comparative evaluation of three molecular typing methods in their applicability to differentiate Lactobacillus strains with human origin

摘要: We isolated a total of 49 strains lactic acid bacteria from the faeces healthy donors. The species in that group were determined as L. plantarum (11 strains), casei rhamnosus (seven fermentum gasseri (six delbrueckii ssp. lactis (four salivarius (two and acidophilus (one strain). Genotyping at strain level was performed using random amplification polymorphic DNA (RAPD), pulsed field gel electrophoresis (PFGE) with endonucleases ApaI XhoI amplified fragment length polymorphism (AFLP) enzymes TaqI. main objective comparison three molecular typing techniques: AFLP, PFGE RAPD their applicability to determine genetic diversity among isolates. easiest, comparatively rapid fairly discriminative tool. most laborious method but producing stable profiles satisfactory discriminatory power. AFLP proved be approach for strains. could differentiate same profiles. Therefore, successfully replace labor consuming PFGE. specially developed very high potential evaluate Lactobacillus spp. human origin.
