Anticonvulsant active inhibitor of GABA transporter subtype 1, tiagabine, with activity in mouse models of anxiety, pain and depression

摘要: Abstract Background Tiagabine, a selective inhibitor of GABA transporter subtype 1 is used as an add-on therapy partial seizures in humans but its mechanism action suggests other potential medical indications for this drug. In research we assess pharmacological activity several screening models seizures, pain, anxiety and depression mice. Methods For tests tiagabine was administered intraperitoneally 60 min before the assay. Behavioral were performed using chemically electrically induced thermal acute pain formalin-induced tonic pain. Anxiolytic-like properties evaluated four plate test elevated plus maze test. Antidepressant-like assessed forced swim addition, to exclude false positive results these assays, influence on animals’ locomotor motor coordination investigated, too. Results Tiagabine demonstrated anticonvulsant (pentylenetetrazole pilocarpine seizures). At dose 100 mg/kg it also seizure threshold by 31.6% ( p Conclusions The obtained present study suggest that tiagabine, apart effect, has anxiolytic-like, sedative antidepressant-like properties. view this, can be potentially treatment mood disorders.
