摘要: Publisher Summary This chapter discusses the isolation and characterization of d-allobisnorbiotin. A pseudomonad is grown aerobically for 5 days on essential-salts medium containing d-biotin as a sole source carbon, nitrogen, sulfur. Double-labeled biotin, thiolane ring, side chain with 3 H ureido carbonyl 14 C are provided in medium, because catabolite loses two-carbon fragment from most original carbon. The culture filtrates concentrated freed soluble proteins by mixing 95% alcohol. separated two anion-exchange column chromatographies Dowex 1 (formate) using ammonium formate an eluent first chromatography formic acid second. catabolite, d-allobisnorbiotin, has trans-ureido ring that very unstable acidic solutions.
