摘要: Electropermeabilization (EPN), also termed electroporation, is a physical method to overcome the barrier of cell membrane by applying short and intense electric pulses. It basis for new cancer treatment modality, electrochemotherapy, where uptake chemotherapeutics enhanced EPN. Preclinical clinical trials have shown that application pulses in vivo feasible electrochemotherapy highly efficient. The aim this study was develop an improved screening drugs on electropermeabilized versus non-electropermeabilized cells. In we describe easy protocol which gives high viability, good reproducibility rate permeabilization. Cell cytotoxicity simply determined MTT assay. death due EPN procedure less than 4% more 90% cells were permeabilized. For daunorubicin, doxorubicin, etoposide paclitaxel, no effect found. carboplatin cisplatin factor 3 2.3, respectively, IC50 (inhibitory concentration 50%). bleomycin found dramatic magnitude 300 IC50. conclusion, established new, reliable test with or without limitations membrane. Our data support role as drug choice electrochemotherapy.
