摘要: A large proportion of Ascobolus immersus transformants are highly unstable in crosses: the phenotype conferred by transgene is not transmitted to progeny, irrespective endogenous or foreign origin transgene. They all have integrated multiple copies, clustered at a single chromosomal site tightly-linked sites. Clustered non-homologous integrations always rearranged. Yet they never escape “methylation induced premeiotically” (MIP) process. This results gene silencing, even when partially repeated, accounting for high instability these transformants.
