Analysis of bacterial communities in soil by PCR–DGGE targeting protease genes

摘要: We studied the effects of application organic (OM) and inorganic fertilizer (CF) on soil protease activity proteolytic bacterial communities in rhizosphere bulk an experimental lettuce field Hokkaido, Japan. The always was higher soils OM than with CF treatment, also soil. analyzed by denaturing gradient gel electrophoresis (DGGE) alkaline metalloprotease (apr) neutral (npr) genes. Most apr forms detected were closely related to Pseudomonas fluorescens, all npr variants resembled gene Bacillus megaterium. These results consistent findings from tests using cultured communities, indicating a high specificity our PCR-DGGE for amplifying community compositions bacteria assessed principal component analysis DGGE profiles. There significant differences apr- npr-expressing bacteria, two types played different roles found correlations between bacteria. indicate that release amounts or activities protease, composition may play major role determining overall activity.
