Epigenetics of multiple myeloma after treatment with cytostatics and gamma radiation

摘要: Genetic and epigenetic changes in multiple myeloma (MM) correlate with the stage of disease. Therefore, we investigated how cytostatics gamma radiation influence MM-associated histone modifications. ChIP-PCR ChIP-on-chip technologies were used to quantify H3K9 acetylation dimethylation at select loci MM patients, lymphoblastoid ARH-77, MOLP-8 cells. Genome-wide analysis revealed that cytostatic, melphalan, increased gene promoters ARH-77 Melphalan also influenced modification prognostically important c-myc CCND1 genes Moreover, was individual patients after melphalan treatment. Western blotting these effects accompanied by c-MYC cyclin D1 protein levels. Taken together, showed significantly alter tumor-related which is indispensable for understanding cancer therapies.
